I will edit the question to include this information. Dials Mavis, it affects Firefox and Safari, but just resetting the cache fixes it for those browsers.

I can create a new cert using our local CA server but I am not sure if this going to make any difference or not? If the page just times out, it has nothing to do with the certificate. That's more to do with port being blocked, or maybe the server isn't listening for traffic on If IIS was listening onbut the cert is wrong, your browser would show you a certificate warning - but a timeout clearly is the missing listener.
To fix it I had to go to the non-working server, and switch the binding to a static cert, then back to the centralised SSL again. Or just recreate the binding from scratch.
Not related to the issue, but I'd also recommend using a 3rd party SSL cert rather than generating your own. Saves a whole world of pain. So a firewall issue somewhere then. Might be the server's Windows firewall, or maybe a "physical" or virtual firewall - or similar. Is there a load balancer in the mix somewhere - or how are you splitting traffic?
If so is port configured on there and does it see both servers as active? Either the IP should be listed or nothing should be listed. As someone mentioned earlier, use the hosts file on the local server to set up site. Then you can test that http and https for site. Remember that https will expect a hostname to match against. If everything works fine on the local server, then you need to work out what sits in between the server and your client that breaks it - or you may find it all works again.
